Run repair permissions

Applications>Utilities>Disk Utilities
Click on your HD in the left column and then repair permissions

EDIT: then log out and back in

Cheers

If you go to System Preferences > Sound, what does it say for "Output volume"? Is the Mute box checked?

That answers one of Van's questions. The sound isn't muted but what is the output set to? Try making sure that it is set to Internal speakers/Built In Output. If it is already set there try setting it to something else for a moment.

No. To be extra "safe" select Archive and Install.

You would lose all of your data if you wiped the drive clean and then installed. If you reinstall, make a backup of all of your data first.

Let's not get hasty though - reformatting is a drastic solution to what is a small problem. Did you check what Slydude suggested?
