So I have had an external hard drive on my imac for a while now, and within the past few months airport lost the connection and I haven't been able to back anything up. So I googled it and read something about partitioning the drive in order for the backup to work.

So, I did. But here's the problem. I selected a drive to partition (really have no clue what I did exactly) then clicked apply. Now my hard drive icon is grayed out on my desktop..and like an idiot I tried restarting and now my computer is stuck on the blue screen with the colored wheel.

What do I do????

It sounds like you may have partitioned the hard drive in your iMac instead of the external drive. How did you do the partitioning? In order to partition the hard drive in your iMac you would have had to boot from your install DVD and run Disk Utility. Please explain exactly what you did so we can recommend a way to get you back up and running again.
