I use TM on an external USB 2TB drive connected to the MacMini but I also have a MacBookPro.
Could I disconnect the USB TM drive and plug it into the MacBookPro and make the occasional full system backup of the MacBookPro then, return the drive to the MacMini.
Would there be two completely separate backups then. One for the MacMini and one for the MBP ?

Yes...

As long as the drive you want to create a TM backup on is formatted properly to begin with, TM does not make any changes to nor touch any existing data on the drive.

My TM backup drive contains the backups for 3 Macs.
I also maintain bootable clones for 2 of the machines that have data I do not wish to lose.
