Use Disk Utility to run a verify and repair on the entire hard drive. Post the results here.

Boot from the install disc by holding down 'C' after the chime, leave Installer and go to Utilities and run Disk Utility > Repair Disk from there and see what is reported. You may have a failed hard drive.

Of course you do have a backup of all your files in case you have to install a new drive?

It sounds like the hard drive has failed. Probably time to swap it out for a new one. Since you have no backup, you may have lost all your data. After removing the hard drive from your iMac, you can place it in an external carrier or use an adapter to see if you can extract your data from it. You'll need a program that can access the drive. Possibly, Disk Warrior may be able to get some or all of your data off the drive. Disk Warrior

By backup was not referring to a disc but an external hard drive with your operating system, folders etc cloned to it. Do you have the install discs for whatever operating system you are running?
