So, if I have a 500GB external HD and I want to run time machine, does that mean if I have 100GB's worth of info on my would my external be filled in 5 hours if I run backup's every hour?

No,
Your time machine only backs up new files that are added. Your first back up is going to be the largest but after that it just backs up the new files that you add to you computer.
