Title says it all: Does my iMac have to be powered on for 'Back to my Mac' to work? Or will it automatically wake up when it receives a request for the 'Back to my Mac' service?

Additional info: my imac would be part of a home wireless network system. and it would nearly always be sleeping rather than shut down-ed.

Be sure Wake for Administrator is enabled in the Power system preference.

But in the preferences, it only says "wake for ethernet network administrator access"

Sorry, I should've clarified that I would be trying to wake the iMac by trying to connect wirelessly, i.e. NOT using an ethernet cable. So if I was in a seperate room and was trying to wake the iMac through my home wireless network, is this possible without using an ethernet cable?

I don't know.
However I suspect the answer is no, due to the fact that the wireless card turns off when the iMac sleeps. That means it will not receive a signal to wake it.
Sorry to rain on your parade.

Just to confirm: yes, your computer does have to be on for Back to My Mac to work?

Which begs the question, do you normally leave your desktop machine awake when you go to work/out?

You can sleep the display without sleeping the HD.
