After I updated to Mac OS X 10.5.2 I discovered that I can no longer change the sound volume level and get a little crossed out circle under my little pop up volume screen.

Can anyone help me fix this problem?

Am assuming you have nothing plugged into the headphone port.

If you do, unplug it. Does that fix it?

If you do not, look in the headphone port. Is there a red light coming out of it? If so, the little thingie in there that turns on when you have an optical cable plugged in is stuck. If this is the case, you can try tickling it with a toothpick to turn off the optical out.

you may be in safe boot mode

If you boot the computer into safe mode (holding down the shift while restarting), then all audio input and output devices will be disabled -- including the internal speakers.

If this is the problem, normal boot should fix it.
