This has been one of my biggest problems with iTunes. Its inability to create a "Now Playing" playlist.

So as a temporary solution, if I want just the one track to play, I just type "apsd9f8i7" in the search box, that is, any random characters which clears the entire playlist and makes sure the track stops after playing...

Just play the song file in quicktime. Then it'll play the song then stop. Not exactly an itunes solution. But it works for me.
