How do I enable Airport Express to link with my MacBook Pro when playing Spotify? It works fine with iTunes and
OSX 10.6.8

Outside of iTunes you would need something like Airfoil from Rogue.

I have looked at Airfoil now - looks like the OS level required is beyond my 10.6 though. Upgrades needed then if I am going to listen to streamed music through my audio system?

Yes, according to the Rogue site you need to be running at least Lion OS X 10.7.X.
