What format is the ringtone in now? Does it play properly for you?

What do you mean by "add to mail"? Do you mean you want to send it to someone?

If you are trying to add a new sound for Mail to use for incoming mail the sound needs to be in an uncompressed (aiff) format. you can convert the sound from other formats using iTunes. The sound also needs to be in specific locations. Here a good overview which mentions several alternatives for converting the sounds.

Here's how to convert sounds in iTunes 11. There's a ton of other software out there, some of it free, that can do this though.
