Manually I'd look to your iTunes library, iPhoto library, documents, downloads and video folders.
You could also use a tool like Disk Inventory X to visualise your disk usage
But be mindful, with great power etc . . . . .
Only delete data that you know is yours. There have been many many horror stories posted where people decide to start pruning files and end up compromising the OS. Then we're into recovery territory when it could all be prevented by making better informed decisions from the off.
