I have several large tiff files from scanned negatives which have been on my hard drive over a year. If I open a file in Preview, rotate it 90 degrees and save it, the entire image turns black!

What OS are you running on what Mac ??
If your on the latest, open in Preview, rotate, then goto the File menu in menu bar click it and then hold down the Option key and 'Save As' will appear, so click that.
Now you get a big dropdown window come up on the Preview window and down the bottom of that it has a box "Save As" and gives you a variety of options. What happens when you save as .jpeg ??
