I'm trying to repair my 13" MacBook Pro (2009) HD with DU off Snow Leopard DVD. The first time I did repair it said it was successful, but I still couldn't boot up off the hard drive. I ran repair again and now I keep getting error message: "could not unmount disk". Is there a way I can still try to repair or is reinstalling OS inevitable? I do have backup.

It sounds like the hard drive may be damaged or failing. You may be faced with changing it out. Since you can't boot from it and now DU refuses to dismount the drive is a good indication that something is wrong with it. Reinstallation of OS X is probably not going to help.
