This is a common problem. I am a Mac owner (MBP 17"). I also work at a company that owns multiple websites. One of our websites takes user submitted videos. Working with this site, I REPEATEDLY see the iSight issue (which I have experienced myself).
Steps to Replicate:
Use a variety of programs and applications which use the cam. Alternate between iChat, Quicktime, Skype, and sites that have flash interface webcams (such as webcams (dot) com).
Result:
The iSight will eventually stop turning on or will only work with one, or some programs, applications. Sometimes, you can fix by restarting your computer. Some users are unable to fix in this manner (and are restricted to the programs that the iSight is "stuck" on).
Desired Result:
The iSight should be able to work from application to application.
Hypothesis:
I think it's possible that the iSight is failing to release the driver.
