Dreamweaver cs4 css how to link to div layer

Hi everybody!

I want to know if its possible to link content to a div layer and if yes how? Let say I have a page with a header DIV, a link DIV and a main DIV, I want the link content from the link DIV to open in the main DIV. I know I can kind of achieve that by creating new html that content the same header and link DIV and change the main DIV, but I think it could be interesting to try to link it directly to the main DIV layer.
