It's a hardware limitation - the first generation MacBook Pro (with the Core Duo CPU) is limited to 2GB total. I don't know if the machine would boot with 4GB installed, but it certainly wouldn't address it. Your best bet would be to use two 1GB matching modules so that you can extract maximum performance from your system.
