I have purchased a new hard drive for the primary purpose of backing up my iMac using carbon clone and possibly a secondary partition set up for video editing.

My questions are
how do I format this hard drive
how do I partition it
I currently have 256 GB free out of 320 GB on my primary hard drive.
After I do this back up with carbon clone, I intend to upgrade to the new operating system( Mountain lion).

Hook the drive up either USB or Firewire, go into Disk utility, select the drive from the left hand pane, hit Erase and choose Mac OS Extended (Journaled) to format. Then into Partition, make sure GUID is selected, and partition as you wish.

One thing Mountain Lion is not the new operating system, Mavericks is. You will have trouble accessing ML now Mavericks has been released.
