I`m over in Japan with a friend. She got a DVD from the States
(region 1 in DVD terms), and to watch it alternated the region setting
on her Japanese Mac book pro. Apparently if you do this more than 5
times it locks the ability to alternate -
MacBook-fr - DVD : Region Free (EN).

Does anyone know if it is possible to reset the region setting?

I don't believe there is. But the free VLC is one way to get around it. It doesn't care what region the disc is.

Currently running OS X 10.8.2

As member Stretch pointed out there isn't a way to alter the Region settings since it's burned into firmware. However, I usually recommend an inexpensive external DVD player which can be used to play "other region" media. That saves the internal drive.

Also, I have found that sometimes even VLC can not get around region settings. It depends on the media.
