iMac - N00b Question

After you download a file from online, what do you do with the .dmg icon that is left on the desktop?
Also, There's 215 Gb's left on my hard drive, I myself haven't installed anything over 10 mb, so did the mac really come with 35 Gb's of software?

If you have already installed the software you got from the .dmg, you can just eject it and delete it.

Even after I install the program, if I try to empty the trash with the .dmg file in it, it tells me I can't because it is in use.

You have to eject the disk image first, THEN you can delet the .dmg file.
